PURPOSE: To enable the stabilization of the subject enzyme for a long period even in the absence of an enzyme-binding substrate by treating a 1- methylhydantoinase according to a specific method and the accurate determination of creatinine or the like.
CONSTITUTION: 1-Methylhydantoinase(NMHase) is treated with (A) a divalent metal ion (preferably Mg+2), (B) a nucleoside triphosphate (preferably ATP) and (C) a complexing agent capable of binding to the component A and forming a complex (preferably ethylenediaminetetraacetic acid). Specifically, the component A is preferably added to an aqueous solution of the NMHase buffered to pH7.5-8.5 and a mixture of the components B with C is then preferably added thereto. 2-[(Tris-hydroxymethyl)methyl]amino-ethanesulfonic acid is preferably added in order to buffer the aqueous solution of the NMHase.
