Abstract:
Provided is an α-amylase which shows an excellent starch decomposition activity at lower temperatures and also shows excellent stability at a low pH value and/or in the presence of a chelating agent. Provided is an α-amylase variant which is a variant of a parent α-amylase, the α-amylase variant including the amino acid residue substitution at a position corresponding to H238-position or E185-positin in the amino acid sequence represented by SEQ ID NO: 2 and the amino acid residue substitution mentioned in (A) or (B) below, in which the parent α-amylase or the α-amylase variant has at least 80% sequence identity to the amino acid sequence represented by SEQ ID NO: 2 (excluding an α-amylase variant having amino acid residue substitutions at three positions corresponding to H238-position, S239-position and G178-position in the aforementioned amino acid sequence, an α-amylase variant having amino acid residue substitutions at three positions corresponding to H238-position, R209-position and G178-position in the aforementioned amino acid sequence, and an α-amylase variant having amino acid residue substitutions at three positions corresponding to E185-position, N190-position and G178-position in the aforementioned amino acid sequence). (A) The amino acid residue substitution at at least one position selected from positions respectively corresponding to T116-position, A181-position, A199-position, A275-position, V277-position, A286-position and L323-position in the amino acid sequence represented by SEQ ID NO: 2; and (B) the amino acid residue substitution at a position corresponding to G178-position in the amino acid sequence represented by SEQ ID NO: 2 and the amino acid residue substitution at at least one position selected from N126-position, T129-position, N190-position, R209-position and S239-position in the aforementioned amino acid sequence.
