Abstract:
Provided are a baculovirus vector and use thereof in the preparation of rAAV in an insect cell. The baculovirus vector comprises an exogenous gene expression cassette and a stable sequence, the stable sequence being located within 5 kb away from the exogenous gene expression cassette, and the stable sequence being a CNE sequence or an NAE sequence. The constructed recombinant baculovirus infects the insect cell, and after successive generations, the production level of the recombinant baculovirus and the rAAV can still be kept relatively stable.
