Abstract:
The present invention provides a carboxyl magnetic bead coupling-modified nucleic acid probe, a preparation method therefor and an application thereof. The preparation method comprises: activating carboxyl magnetic beads to obtain activated carboxyl magnetic beads; and co-incubating the activated carboxyl magnetic beads and an amino-modified nucleic acid probe in a coupling reaction solution to obtain the carboxyl magnetic bead coupling-modified nucleic acid probe. The coupling reaction solution is a mixed solution of morpholine ethanesulfonic acid and N,N-dimethyl acetamide. The carboxyl magnetic bead coupling-modified nucleic acid probe is used for targeted capture of target nucleic acid in a biological sample. The nucleic acid obtained by targeted capture can satisfy requirements of gene mutation detection and gene methylation detection, and can avoid influence of a background gene and an inhibitor in the biological sample with high capture efficiency. The present invention can perform targeted capture of specific genes in low-abundance DNA/RNA samples such as blood, urine, and feces.
