Provided is a cell culture method with which, in cell culturing using a culture vessel, it is possible to suitably control the exchange rate of a medium in the vessel. This cell culture method uses: a culture vessel which is filled with cells and a medium and in which cell culture is carried out; and a medium supplying vessel which is filled with a new medium and which supplies the medium to the culture vessel. The culture vessel has at least a first port which is used for medium supply, and a second port which is used for medium discharge. The medium supplying vessel serves as a mixing vessel for adjusting the concentration of a new medium to be supplied to the culture vessel. A portion of used medium in the culture vessel is sent from the culture vessel to the mixing vessel. In the mixing vessel, a new medium and the portion of used medium are mixed to prepare a concentration-adjusted medium. While the concentration-adjusted medium is transferred from the mixing vessel to the culture vessel, the remaining used medium is discharged from the culture vessel, and the culture vessel is filled with the concentration-adjusted medium.
