Disclosed is a cell sorting system based on endonuclease specific recognition. The sorting system comprises the following components: a first single nucleotide, a second single nucleotide, an antibody, a cell separation medium and a restriction endonuclease. The first single nucleotide and the second single nucleotide are complementary, and a complementary area comprises a recognition site of the restriction endonuclease. The first single nucleotide, the second single nucleotide, the cell and the cell separation medium are all modified, the modified first single nucleotide may connect to the modified antibody, and the modified second single nucleotide may connect to the modified cell separation medium. Further disclosed are a method for sorting out a cell by using the system and applications thereof, and a kit and method thereof for sorting out a DC cell, an NK cell and a CIK cell from a cell sample.
