Abstract:
Disclosed are a chimeric antigen receptor, a macrophage expressing same, and use. A primary protein structure of the chimeric antigen receptor comprises sequentially from the amino terminus to the carboxyl terminus: a signal peptide, an NKG2D extracellular region, a hinge region, a transmembrane region, and an intracellular signal region; the NKG2D extracellular region is a human NKG2D extracellular domain, an amino acid sequence thereof being set forth in SEQ ID NO. 2. The chimeric antigen receptor macrophage of the present invention comprises an encoding gene of the chimeric antigen receptor or an expression vector of the encoding gene of the chimeric antigen receptor. In addition to being capable of directly killing tumor cells, the chimeric antigen receptor macrophage of the present invention has the advantages of enabling improvement of a solid tumor immune microenvironment, being able to present an antigen as an antigen-presenting cell after phagocytizing tumor cells, infiltrating into tumors more easily and cooperating with other immune cells to infiltrate into tumors, etc.
