Abstract:
Provided are a Chlamydomonas reinhardtii chloroplast-Saccharomyces cerevisiae-Escherichia coli shuttle vector, a method for constructing same, and use thereof. The construction method comprises design, chemical synthesis, assembly and identification of the shuttle vector and replication of the shuttle vector in the Escherichia coli. The constructed shuttle vector can shuttle and replicate between the Escherichia coli, yeast and Chlamydomonas reinhardtii chloroplast. The shuttle vector comprises a replication element (CEN) of the yeast, a replication element (ori) of the Escherichia coli, and a Chlamydomonas reinhardtii chloroplast genome (SynCp V1.0). Chlamydomonas reinhardtii cells are transformed with the shuttle vector, thereby achieving the biological function of the shuttle vector; the chloroplast genome is extracted and transferred into the Escherichia coli, thereby achieving successful replication of the shuttle vector in the Escherichia coli; and the shuttle vector capable of shuttling and replicating between the Escherichia coli, yeast and Chlamydomonas reinhardtii chloroplast is constructed for the first time.
