Abstract:
Provided are a construction method and use of an astaxanthin synthesis pathway in Chlamydomonas reinhardtii. The construction method comprises design and assembly of an astaxanthin synthesis module and functional verification of the astaxanthin synthesis module in Chlamydomonas reinhardtii. Seven genes related to astaxanthin synthesis are selected, and an astaxanthin expression module framework is designed and divided into four fragments for artificial synthesis; then the four fragments and a yeast-E. coli shuttle plasmid backbone are co-transfected into a yeast to obtain a yeast containing the astaxanthin synthesis module; then the yeast containing the astaxanthin synthesis module is hybridized with a yeast containing a chloroplast genome to insert the astaxanthin synthesis module into a Chlamydomonas reinhardtii chloroplast genome, a recombinant plasmid is introduced into the Chlamydomonas reinhardtii chloroplast, and finally a positive Chlamydomonas reinhardtii transformant containing the astaxanthin synthesis module is obtained, namely an astaxanthin synthesis engineering algae strain. The finally obtained transgenic astaxanthin synthetic engineering algae strain can realize rapid and cheap production of astaxanthin by means of large-scale culturing.
