Abstract:
The present invention relates to an antibody specific to Glypican 3 (GPC3), and a use thereof and, more specifically, to: an antibody specifically binding to GPC3; a chimeric antigen receptor comprising the antibody; a CAR-T cell expressing the chimeric antigen receptor; and a pharmaceutical composition for preventing or treating diseases mediated by GPC3-expressing cells, comprising the antibody, the receptor, and the cell. In the present invention, six novel types of antibodies (10E9, 8B2, 7C6, 7B9, 5E11, 1A8) were established by screening for antibodies more specifically binding to GPC3, and it has been confirmed that the novel antibodies specifically bind to a GPC3 antigen. Also, it has been confirmed that the GPC3-CAR-T cell produced in the present invention effectively recognizes the GPC3 antigen, and, thereby, activation of the CAR-T cell is induced, and the GPC3-expressing cells are effectively killed. Therefore, the GPC3-specific antibody, the chimeric antigen receptor produced by using same, and the CAR-T cell can be applied for prevention or treatment of cancer or a tumor expressing GPC3.
