Abstract:
To provide a preparation method of a new sample which, without requiring a definite diagnosis using a biopsy, can be used as a liquid biopsy sample in prenatal diagnosis of a fetus. This isolation method of fetal-derived hematopoietic progenitor cells is characterized by involving an emulsion forming step, a culturing step, a detection step and a selection step. In the emulsion forming step, a blood sample of the pregnant woman, a culture medium, and an emulsion forming solvent are brought into contact to form, in the emulsion forming solvent, multiple droplets that contain the pregnant woman-derived hematopoietic progenitor cells or fetus-derived hematopoietic progenitor cells contained in the blood sample. In the culturing step, the aforementioned multiple droplets are cultured in the presence of a growth factor of hematopoietic progenitor cells. In the detection step, the fetus-derived hematopoietic progenitor cells are detected in the droplets after culturing. In the selection step, droplets in which the fetus-derived hematopoietic progenitor cells have been detected are selected from the droplets after culturing.
