Abstract:
Disclosed is a novel molecular target through which aspirin exerts a neuroprotective effect on cerebral apoplexy. Aspirin exerts a neuroprotective effect on a mouse model of cerebral apoplexy by means of acetylating calcium ion-calmodulin dependent protein kinase 2α (CaMK2α) within neurons. The present invention describes a novel mechanism through which aspirin exerts a neuroprotective effect independent of the anticoagulant function thereof. Further disclosed is that an aspirin derivative may also exert a neuroprotective effect in a mouse model of cerebral apoplexy by means of acetylating CaMK2α; however, the anticoagulant effects and bleeding risks thereof are weaker than the anticoagulant effects and bleeding risks of aspirin. Further disclosed is a novel strategy for safely and effectively preventing cerebral apoplexy: by means of modifying aspirin, anticoagulant activity thereof is reduced while retaining the acetylation effect thereof on CaMK2α, which may result in a better neuroprotective effect in preventing cerebral apoplexy and reduce the side effect of bleeding. The present invention provides a safer and more effective approach for prophylactic medication against cerebral ischemic diseases.
