Abstract:
Provided is a fluorescence–patch clamp–micropipette measurement apparatus. An experiment platform (31) is equipped with an experiment chamber (22), a three-dimensional micromanipulator (23, 24), and a patch clamp; two sides of the experiment chamber (22) are hollowed out for a micropipette (34) and a glass electrode (25) to enter; the experiment chamber (22) has fluorescent cells (32); the micropipette (34) sucks red blood cells (33); the cells (32) and red blood cells (33) are in the extracellular fluid in the experiment chamber (22); the glass electrode (25) is filled with electrode inner liquid (30); the patch clamp comprises a patch clamp amplifier (20), a patch clamp probe, and a patch clamp probe holder; full-spectrum light (12) emitted by a fluorescent light source (11) passes through a color filter (13) to form fluorescent incident light (14), and is incident on the cells (32) in the experiment chamber (22); the fluorescence return of the cells (32) is received by a fluorescence camera (16). The measurement apparatus can detect the effect of cell membrane potential changes on the interaction of membrane proteins while simultaneously collecting fluorescence images with a high signal-to-noise ratio in order to study transmembrane signal transduction of membrane proteins, and can record three-spectrum phase information such as fluorescence spectrum, electrophysiological spectrum, and adhesion state spectrum while simultaneously recording the coupling relationship between the three-spectrum phase information.
