Abstract:
The problem is to produce and/or maintain naïve pluripotent stem cells capable of high expression of genes important to maintain an undifferentiated state, which could not be achieved by known methods for producing pluripotent stem cells. The problem of the invention can be solved because the invention makes it possible to produce naïve pluripotent stem cells capable of maintaining an undifferentiated state by introducing six genes (Oct3/4, Klf4, c-Myc, Sox2, Nanog, and Klf2) among the so-called reprogramming factors, inducing transient expression as a characteristic feature, and performing culturing in medium containing LIF, MEK inhibitor, GSK3 inhibitor, cAMP production promoter, TGF-β inhibitor, and PKC inhibitor.
