Abstract:
Provided is a method for purifying an antibody-like protein, said method including a step for eluting under weakly acidic conditions. This method for purifying an antibody-like protein contains the following steps (a) and (b): (a) a step in which an antibody-like protein is brought into contact with an affinity separation matrix containing a ligand immobilized on a carrier, and is suctioned to the affinity separation matrix; and (b) a step in which an eluate having a pH of at least 3.5 is brought into contact with the affinity separation matrix, and the antibody-like protein is eluted, wherein the ligand contains an amino acid sequence obtained by replacing, in an amino acid sequence derived from the E, D, A, B, or C domain of a protein A described by SEQ ID NO: 1-5, a Gln and/or Lys of an Fc binding site with an Ala, Ser, and/or Thr, and the ligand has a reduced antibody binding capacity in acidic pH regions in comparison with the pre-replacement ligand.
