Disclosed are polypeptide compounds using PD-1 receptors as targets. The polypeptide compounds comprise a compound 1, a compound 2, a compound 3, a compound 4 and a compound 5. The amino acid sequences of the compounds 1-5 are as shown in SEQ ID NOs: 1-5. Further disclosed is a method for preparing polypeptide compounds using PD-1 receptors as targets, comprising the following processes: sequentially coupling the amino acids of the compounds 1-5 onto a solid-phase resin according to peptide sequences by using a solid-phase synthesis method; and performing cleavage by a cutting reagent, freeze-drying, and preparing and purifying to obtain a target compound. Further disclosed is a use of polypeptide compounds using PD-1 receptors as targets in the preparation of analgesic drugs. The small molecule polypeptides of the present invention, the compounds 1-5, can bind to PD-1, and have a significant inhibitory effect on both inflammatory pain and visceral pain by intrathecal administration; and the synthesis is simple and the purification is easy, facilitating large-scale synthesis.
