Abstract:
The present invention belongs to the biotechnology field of stem cell preservation. Provided are a preservation fluid used for human menstrual blood and a preparation method therefor. The ingredients of the preservation fluid comprise normal saline, hirudin, glucose, hydroxyethyl starch, levofloxacin, multivitamins, glutathione and a necroptosis inhibitor Necrostatin-1/RIP1 kinase inhibitor. The preparation method for the preservation fluid is as follows: 1) preparing a solution A: respectively adding glucose, hirudin and levofloxacin into normal saline; 2) preparing a solution B: respectively adding hydroxyethyl starch, multivitamins and glutathione into the normal saline; and 3) preparing a solution C: mixing the solution A and the solution B, then adding the necroptosis inhibitor Necrostatin-1/RIP1 kinase inhibitor, and after filtering by using a filter membrane, storing at 4°C for later use. The preservation liquid of the present invention may maintain the original biological characteristics of endometrial stem cells within 48 hours, the cell quality is stable, and the purity is high.
