Abstract:
Method for producing a complex of magnetic particles and a disulfide-binding protein in a gram-negative magnetotactic bacterium, the method involving: a coexpression step (i) in which a first fusion protein (a) containing a disulfide-binding protein fused to a binding protein, and a second fusion protein (b) containing a binding partner protein fused to a protein that can bind or integrate with bacterial magnetic particles are coexpressed in a gram-negative magnetotactic bacterium so that the expression of the first fusion protein is induced to the periplasm of said bacterium and the expression of the second fusion protein is induced to the cytoplasm of the aforementioned bacterium; a culture step (ii) in which the bacterium of step (i) is cultured under conditions sufficient for the production of the first and second fusion proteins; and a cell membrane breakdown step (iii) in which the cell membrane of the bacterium of step (ii) is broken down to form a complex of the disulfide-binding protein and bacterial magnetic particles.
