Title:
RECOMBINANT BACTERIUM FOR PRODUCING SHIKIMIC ACID, CONSTRUCTION METHOD THEREFOR, AND APPLICATION THEREOF
Document Type and Number:
WIPO Patent Application WO/2023/092633
Kind Code:
A1
Abstract:
Provided is a recombinant bacterium for producing shikimic acid, for use in expressing a target gene for regulating asymmetric cell division and target genes for regulating shikimic acid production. The target gene for regulating the asymmetric cell division comprises a gene popZ for coding cytoskeleton protein PopZ, and the target genes for regulating shikimic acid production comprise a gene aroG for coding DAHP synthetase, a gene aroB for coding 3-dehydroquinic acid synthase, and a gene tktA for coding transketolase. The recombinant strain implements de novo synthesis of shikimic acid, a substrate is glucose, the cost is low, and after the strain is fermented in a 7.5 L fermentation tank, the highest yield of shikimic acid is 88.1 g/L, the yield is 0.33 g/g, and the production intensity is 1.1 g/L/h.
