Abstract:
Disclosed is a simple and highly efficient method for the induction of a Puerariae lobatae radix polyploid. The method comprises the steps of treating an explant, culturing an aseptic seedling, inducing a cluster bud, culturing for stem and node elongation, inducing a polyploid, separating a chimera, screening a pure line, etc. The method comprises firstly culturing a young vine to obtain an aseptic seedling, then inducing the aseptic seedling to obtain a cluster bud, and further culturing the obtained cluster bud by using a medium to obtain a tissue culture seedling with relatively long stem and node. A single bud stem section with relatively long stem and node can be obtained by means of this method and can also be used as an induction material, enriching the existing materials for inducing a Puerariae lobatae radix polyploid and improving the working efficiency of induction. In addition, the inducing rate of using the single bud stem section of Puerariae lobatae radix as the induction material is higher than that of using a shoot tip.
