Abstract:
The present invention relates to a supramolecular assembly preparation method and a method for displaying a protein on a supramolecular assembly, the supramolecular assembly preparation method comprising the steps of: (a) preparing a plurality of assembly monomers comprising (i) a first split fragment containing green fluorescent protein (GFP) β-strand 1-10 and (ii) a second split fragment containing GFP β-strand 11 coupled to the terminus of the first split fragment; and (b) preparing a supramolecular assembly by carrying out self-assembly of a plurality of the assembly monomers. In addition, the present invention relates to a method for purifying a supramolecular assembly of a polydisperse distribution into a monodisperse supramolecular assembly, comprising the steps of: (a) mutating assembly monomers such that the negative charge value increases by reducing the net charge of the assembly monomers; (b) preparing a negatively charged supramolecular assembly of a polydisperse distribution by carrying out a plurality of self-assemblies of the assembly monomers; (c) separating the supramolecular assembly of a polydisperse distribution according to the size of the assembly; and (d) collecting the separated monodisperse supramolecular assembly.
