Abstract:
A system and a method for instantly detecting nucleic acid of a pathogen. The system comprises a lysis buffer, which is used for inactivating and lysing a sample to be detected, so as to release RNA; an RT pre-mixed solution, which is used for reversely transcribing the RNA in the sample into DNA; an RPA pre-mixed solution, which is used for performing an RPA reaction on the sample treated with the RT pre-mixed solution, so as to realize amplification of the DNA in the sample; a CRISPR reaction pre-mixed solution, which is used for performing a specific cleavage reaction on the sample treated with the RPA pre-mixed solution; and colloidal gold test paper, which is used for performing color development on the sample treated with the CRISPR reaction pre-mixed solution, to confirm whether the sample contains the nucleic acid of the pathogen. According to the above-mentioned design scheme, the RPA method, the CRISPR method and the colloidal gold method are coupled, so that the instant-detection system has the advantages of simplicity, sensitivity and specificity, instruments and equipment are not required, and same can be used to quickly obtain a result by means of simple operation at room temperature.
