Abstract:
PURPOSE: To obtain a DNA compound encoding isopenicillin N:acylCoa acyltransferase activity and giving a gene related to the production of penicillins.
CONSTITUTION: This DNA compound contains an isolated DNA sequence encoding the isopenicillin N:acylCoA acyltransferase activity of Aspergillus nidulans. The DNA compound encoding the acyltransferase activity is isolated from the genome DNA of the Aspecrgillus nidulans. The cloned acyltransferase gene is useful for increasing the yield of penicillin G in Aspergillus or Penicillium, especially when a reaction catalyzed with the acyltransferase is a rate- determining step. This is achieved by increasing the amount of the gene and/or disposing a code region behind a strong promoter.
