Abstract:
PURPOSE: To provide a novel enzyme that hydrolyzes amino-methoxy-methyl- cephem-carboxylic ester to chemoselectively form amino-methoxymethyl-cephem- carboxylic acid in high yield.
CONSTITUTION: This is an esterase that is suitable for the chemoselective conversion of an isomer of (1R,S)-1-(2,2-dimethylpropionyloxy)-ethyl 7-amino-3- methoxymethyl-3-cephem-4-carboxylate of formula I to 7-amino-3-methoxy- methyl-3-cephem-4-carboxylic acid. This enzyme is prepared by culturing a microorganism producing the enzyme, suitably Streptomyces bambergiensis (ATCC 13879), preferably at 28°C and collecting the objective enzyme from the culture mixture through the known process.
