Abstract:
PURPOSE: To provide new tropane derivatives useful for treating drug abuse, depression and Parkinson's disease.
CONSTITUTION: The compounds represented by formulae I or II, e.g. (1R,2R,3 S)-2-(4-chlorophenoxy-methyl)-3-(4-fluorophenyl) tropane. are obtained by subjecting compounds of formula II to a Michaelis 1,4-addition reaction with formula R4-A (A is a reactive functional group). In formulae I or II, R is H, alkyl, alkenyl, alkynyl, 2hydroxyethyl or the like; R3 is (CH2)NCO2R11, COR11, CH2R12; R4 is (substituted) phenyl, (substituted) benzyl or the like; R11 is alkyl, cycloalkyl, (substituted) pylidyl or the like; R12 is (substituted) phenoxy, (substituted) benzoyloxy; (n) is 0 or 1.
