Abstract:

PURPOSE: To enable the production of a large amount of the titled DNA, by integrating simian virus 40DNA devoid of the replication starting point to a specific plasmid, and transforming a host cell with the obtained recombinant plasmid.

CONSTITUTION: A simian virus 40DNA devoid of replication starting point is prepared, and separately, a plasmid proliferable in a host cell and having a restriction enzyme-recognizing site common to the single restriction enzyme- recognizing site existing in said DNA is prepared. The simian virus 40DNA devoid of replication starting point is integrated in the plasmid to obtain a recombinant plasmid proliferable in a host cell. The host cell is transformed by the recombinant plasmid, the transformation is cultured, and the plasmid is recovered from the cultured microbial cell. The simian virus 40DNA devoid of replication starting point is separated from the recovered plasmid.
