Abstract:
Disclosed are cationic lipids for gene transfer, which are capable of transferring nucleic acid materials into cells at very high efficiency with low cell toxicity. They are synthesized by forming a carbamate ester linkage between lysine or ornithine residue and a cholesteryl chloroformate in a solid-phase manner. This solid-phase synthesis method allows the automatic mass production of the vehicles suitable for use in gene therapy and transfer. Further, disclosed are liposomes made of the cationic lipids. These liposomes are very useful in transfecting nucleic materials into cells as well as in delivering pharmaceutically active materials into target cells.
