Abstract:
Disclosed are a genetically engineered strain capable of producing L-alanine, a construction method therefor, and an application thereof. A genetically engineered strain capable of efficiently producing alanine at a high temperature of 42°C to 55°C can be constructed by enhancing a glycolysis pathway or/and introducing a thermostable alanine dehydrogenase gene. The optical purity of L-alanine can further be significantly improved by knocking out an alanine racemase gene. When a lactic acid synthesis pathway exists in starting bacteria, the proportion of a pyruvic acid synthesis pathway can be increased by blocking the lactic acid synthesis pathway, so as to further improve the yield of L-alanine, such that high-temperature fermentation production of L-alanine at 42°C-55°C is achieved, and the yield of alanine can reach more than 95 g/L.
