Abstract:
PROBLEM TO BE SOLVED: To immunologically determine an antigen, antibody, or the like, in high sensitivity by detecting a marker enzyme using a chemiluminescent reagent containing a specific lucigenin derivative in an enzyme immunoassay using a peroxidase as the marker enzyme.
SOLUTION: An antibody or antigen labeled with a peroxidase enzyme is mixed with a determination object antigen or antibody or their coagulate in a specimen to form a peroxidase-labeled antibody-antigen complex by antigen- antibody reaction. The antigen or antibody is quantitatively determined by chemiluminescence using a chemiluminescent reagent in the presence of a hydrogen receptor. The chemiluminescent reagent is prepared by reacting an N,N'- disubstituted-9,9'-bisacridinium salt of formula I [R1 and R2 are each an alkyl or a (substituted) aryl; R3 to R6 are each H, an alkyl, an aryl, a halogen or the like; X is an n-valent anion; (n) is 1 or 2] (e.g. lucigenin) with an N,N- disubstituted carboxylic acid amide of formula II (R1 is H, an alkyl or an aryl; R2 is methyl or ethyl; R3 is an alkyl, an aryl or the like) under light irradiation.
